Currently, it isn’t possible to set up Automations that add or remove labels based on the specific day of the week, such as automatically adding a "Wednesday" label when the due date falls on a Wednesday and removing a "Friday" label. None. However, some users can use a Custom Field instead of a label and create an automation such as:

• when a due date is set on a card, set custom field "Day of the week" to "{cardduedateweekdayname}"
