When you create an issue from Confluence into JIRA, currently you cannot set or assign the created ticket from Confluence side.
Therefore, to set the assignee, it's necessary to go to JIRA, open the newly created issue and then set the assignee from there.

You can try using an add-on like Jcreate that allows you to create a Jira Issue from inside confluence while adding many customisable fields such as assignee.
